The Atg8 family of ubiquitin-like proteins play pivotal roles in autophagy and other processes involving vesicle fusion and transport where the lysosome/vacuole is the end station. Nuclear roles of Atg8 proteins are also emerging. Here, we review the structural and functional features of Atg8 family proteins and their protein-protein interaction modes in model organisms such as yeast, Arabidopsis, C. elegans and Drosophila to humans. Although varying in number of homologs, from one in yeast to seven in humans, and more than ten in some plants, there is a strong evolutionary conservation of structural features and interaction modes. The most prominent interaction mode is between the LC3 interacting region (LIR), also called Atg8 interacting motif (AIM), binding to the LIR docking site (LDS) in Atg8 homologs. There are variants of these motifs like “half-LIRs” and helical LIRs. We discuss details of the binding modes and how selectivity is achieved as well as the role of multivalent LIR-LDS interactions in selective autophagy. A number of LIR-LDS interactions are known to be regulated by phosphorylation. New methods to predict LIR motifs in proteins have emerged that will aid in discovery and analyses. There are also other interaction surfaces than the LDS becoming known where we presently lack detailed structural information, like the N-terminal arm region and the UIM-docking site (UDS). More interaction modes are likely to be discovered in future studies.
The Northern Svalbard Composite Tectono-Sedimentary Element (CTSE) comprises Proterozoic, Early Paleozoic, and Devonian sedimentary rocks preserved in northern Svalbard and on the adjacent shelf margin. Here, the sedimentary strata of the CTSE are preserved between complexes of metamorphic and crystalline basement rocks. The Northern Svalbard CTSE covers four main tectonostratigraphic elements: Tonian syn-rift, Neoproterozoic to Cambrian post-rift, Ordovician passive margin, and late Silurian?/Devonian syn-extensional basins. The oldest documented sedimentary strata are greywacke and shale deposited after the Greenvillian Orogeny and assumed to be younger than 980-960 Ma. The present CTSE further includes Cambrian-Ordovician sedimentary rocks in Ny-Friesland and Nordaustlandet, while the dominant part of the CTSE are continental Old Red Sandstone sediments of the late Silurian?/Early Devonian Red Bay and Siktefjellet groups and the Early-Late Devonian Andreé Land Group onshore northern Spitsbergen. Locally, the Cambrian-Ordovician formations contain petroleum source rocks with moderate total organic carbon contents and relatively high hydrogen index values, suggesting a good potential for oil generation. There are possible reservoirs, seals, and traps in some of the basins, but the CTSE generally holds a very limited potential as a petroleum province, particularly as the region is under strict environmental protection
Ageing civil infrastructures such as bridges and building causes many consequences from practical and economical point of view. Especially in northern Norway, the impact of extreme arctic conditions is intense on civil engineering infrastructures (see \cite{Singh:2022:hologaland_Bridge}). With the increased loading due to the prosperous seafood industry and increased cargo activity is putting additional pressure on the aged infrastructures. Research and development of new methods is needed for the damage detection in these structures. In this paper we present, discuss and analyze the situation concerning bridges in Norway with a special focus on northern Norway. Moreover, based on the research in \cite{singh:2022:phdThesis}, \cite{singh:2019OMA-Application} and \cite{singh:2021:comprehensive} we describe and emphasise the importance of structural health monitoring methods, artificial intelligence and machine learning when trying to solve these serious problems of structural damage detection especially in arctic regions. The related mathematical tools, in particular the role of equation of motion, are discussed. In a certain specific case of the equation of motion there is discussed the case where the solution may be obtained in the explicit form via the use of the so-called Calder\'on operator.

More than half of the population with HIV in the world live in sub-Saharan Africa. HIV-associated chronic lung disease is the most prevalent comorbidity among children and adolescents with HIV in this region. The pathogenesis is multifactorial and thought to be a result of chronic immune activation and repeated lung injury. CD4+ T-cells are the main target for HIV infection. As the majority of CD4+ T-cells reside in the gut, HIV can cause a disrupted gut barrier and gut microbial dysbiosis, contributing to maintain systemic immune activation. We used data from a randomized, placebo-controlled trial of 347 children and adolescents with HIV-associated chronic lung disease in Zimbabwe and Malawi, investigating the effect of azithromycin on lung function. Rectal swabs were collected at inclusion, after 48 weeks of treatment and 6 months after cessation of study drug. 16S rRNA sequencing was performed to assess the gut microbiota. The overall aim of this thesis was to investigate the the gut microbiota of children and adolescents with HIV, and to study the effects of antibiotic (azithromycin) treatment on gut microbial composition and on local lung inflammation. We found that participants with HIV had a reduced gut microbial diversity compared to participants without HIV. Diversity improved with longer time on antiretroviral therapy. Azithromycin reduced the overall gut microbial diversity, but these changes did not persist 6 months after cessation of antibiotics. Exhaled nitric oxide, a measure of local inflammation in the lungs, was associated with acute respiratory exacerbations and biomarkers of inflammation. We found no effect of azithromycin on exhaled nitric oxide. This work highlights the association between HIV and gut microbial dysbiosis and the effect of long-term antibiotics on lung inflammation and composition of gut microbiota. It adds knowledge to the field of HIV-associated comorbidities and can be helpful when designing new studies on gut microbiota and potential microbiota-related targets for interventions in the future.; Over halvparten av de som lever med HIV i verden bor i Afrika, sør for Sahara.
UiT Norges arktiske universitetUiT The Arctic University of NorwayLundgren, Ingela MarieMastergradsoppgaveMaster thesisEstablishment of killer whale cell cultures and their responses to pollutant exposureBjørneset, Junihttps://hdl.handle.net/10037/331732024-03-15T07:21:04Z2022-03-15T00:00:00ZBjørneset, Juni
The cosmopolitan apex predator killer whale (Orcinus orca) is one of the species worldwide with the highest levels of persistent organic pollutants (POPs) in their tissues. The knowledge of how POPs may affect the species is limited, likely due to the ethical, legal, and practical challenges of researching toxicology on free-ranging marine mammals. The use of in vitro models has proved to be a functional tool which may provide information of cellular responses to POPs exposure in marine mammals. Skin biopsies of killer whales were sampled from the Northern Norwegian fjords during aggregation of killer whales for foraging on spring spawning herring. Primary fibroblast-like cells were established from the dermis of 11 out of 13 killer whales sampled. Cytotoxic and gene transcript analysis were conducted on the fibroblast-like cells exposed to ecologically relevant concentrations of POPs, reflecting the 10 most abundant POPs found in Norwegian killer whales. The pollutants were applied at different concentrations to explore dose-dependent responses. Significant downregulation of the target gene CYP1A was observed at medium and highest exposure concentration, as well as non-significant tendencies of another downregulated gene (CD36) and seven upregulated genes (ADIPOQ, CYP4A, ERA, GR, PPARA, PPARG and THRA) at the highest concentration of POPs exposure. This study provides a successful establishment of killer whale fibroblast-like cells, as well as proven gene alterations of the cells exposed to POPs.
We consider linear codes over a finite field Fq, for odd q, derived from determinantal varieties, obtained from symmetric matrices of bounded ranks. A formula for the weight of a codeword is derived. Using this formula, we have computed the minimum distance for the codes corresponding to matrices upperbounded by any fixed, even rank. A conjecture is proposed for the cases where the upper bound is odd. At the end of the article, tables for the weights of these codes, for spaces of symmetric matrices up to order 5, are given.
We study varieties associated to hypergraphs from the point of view of projective geometry and matroid theory. We describe their decompositions into matroid varieties, which may be reducible and can have arbitrary singularities by the Mnëv–Sturmfels universality theorem. We focus on various families of hypergraph varieties for which we explicitly compute an irredundant irreducible decomposition. Our main findings in this direction are three-fold: (1) we describe minimal matroids of such hypergraphs; (2) we prove that the varieties of these matroids are irreducible and their union is the hypergraph variety; and (3) we show that every such matroid is realizable over real numbers. As corollaries, we give conceptual decompositions of various, previously studied, varieties associated with graphs, hypergraphs, and adjacent minors of generic matrices. In particular, our decomposition strategy gives immediate matroid interpretations of the irreducible components of multiple families of varieties associated to conditional independence models in statistical theory and unravels their symmetric structures which hugely simplifies the computations.
