dc.description.abstractViral infections are a leading cause of mortality in the farming of Atlantic salmon in Norway. Viral outbreaks cause heavy losses each year, and one of the most predominant viral diseases is infectious pancreatic necrosis (IPN). The virus causing this disease, the infectious pancreatic necrosis virus (IPNV), belongs to the Birnaviridae family of viruses with a double-stranded RNA (dsRNA) genome. Although viral disease is prevalent in the Atlantic salmon aquaculture industry, fish possess an immune system fully equipped to handle viral disease. In this work we have studied induction of the type I interferon (IFN) response in Atlantic salmon in response to intracellular foreign RNA. IFNs are pivotal alarm proteins alerting other cells of the infection, contributing to the establishment of an antiviral state. In order for IFNs to be produced, the cell must recognize the presence of the viral intruder. Viruses are recognized by a wide range of different receptors. The receptors recognize characteristic pathogen associated molecular patterns (PAMPs), for viruses usually the nucleic acids. The receptors proceed to signal through a conserved signaling pathway that culminates in the induction of IFN production. We have identified a central member of the signaling pathway, IPS-1, a signal mediator conserved through evolution. We have also investigated the promoter elements of two type I IFNa promoters, characterizing the regulatory regions. The induction of IFN production is a common target for viral proteins trying to subvert the IFN response and subsequent antiviral state. Using the IFN promoters and IPS-1 protein, we identified IFN antagonistic effects of separate IPNV proteins.en

dc.description.popularabstractVirusinfeksjoner er årsaken til store økonomiske tap i oppdrettsnæringen i store deler av verden. En av virussykdommene som er hyppigst forekommende i Norge er infeksiøs pankreas nekrose (IPN). Viruset som forårsaker denne sykdommen, infeksiøs pankreas nekrose virus (IPNV) tilhører Birnaviridae familien av virus, og har et totrådig RNA genom. Det er viktig å kartlegge hvordan laks forsvarer seg mot virusinfeksjoner. Til tross for at flesteparten av oppdrettslaksen i Norge er vaksinert mot IPNV er det fremdeles hyppige IPN utbrudd. Til tross for de hyppige virusutbruddene i akvakulturindustrien så har fisk, deriblant laks, et immunsystem som er fullt utrustet til å takle virussykdommer. I dette arbeidet har vi studert induksjon av type I interferon (IFN) responsen hos atlantisk laks. IFN er essensielle alarmproteiner som kan fortelle andre celler at det er en infeksjon. Når IFN signalerer til celler, kan disse cellene etablere en antiviral tilstand, som kan forhindre videre utvikling av infeksjonen. For at IFN skal bli produsert må de infiserte cellene oppdage at det er et virus til stede. Virus kan kjennes igjen av en rekke ulike reseptorer. Reseptorene kjenner igjen karakteristiske molekylære trekk ved patogene organismer, for virus er det vanligvis nukleinsyrene som utgjør virusgenomet som blir gjenkjent. Når reseptorene har blitt aktiverte, sender de signalene videre via en signalvei som er konservert gjennom evolusjonen. Signalveien ender i at IFN blir produsert. Vi har identifisert at også laks har et sentralt medlem av signalveien, IPS-1. Vi har også undersøkt promoterelementene som styrer produksjon av IFN i to type I IFNa promotere. Induksjon av IFN produksjon er et vanlig mål for virusproteiner som forsøker å hindre IFN responsen, og dermed hindre at det blir etablert en antiviral tilstand. Slike proteiner kalles IFN antagonister. Vi undersøkte IFN antagonistiske effekter av de separate IPNV proteinene, og fant at flere av proteinene hadde kraftige mekanismer for å forhindre produksjon av IFN.en
